It has a neon in it. A neon works on a tiny amount of current. The screwdriver has a metal top at the back, and the neon sits between that and the tip of the screwdriver. If you press the tip against mains electricity and press your thumb against the metal top, a tiny current passes through the neon and you. It's not enough for you to notice, but it is enough to light the neon.

how does it allow you to distinguish between the live and neutral wire? does it glow a different colour?

It doesn't. It merely glows if you touch the blade onto a live 220v wire with your thumb or finger on the ferrule at the top of the handle. This is usually used to ensure that the power is disconnected from what you are working on.

What type of batteries does this take, as mine didn't come with any

It doesn’t need batteries. When you touch the blade on the conductor you’re testing, with a finger on the handle cap, you create a path for current to flow from the conductor, through a neon lamp, and into your finger, causing the lamp to illuminate if a voltage is present. Needless to say, the current is very small and harmless.
